【编者按】执业药师综合辅导:中药脱色方法
目前应用于中药脱色的方法及工艺很多,但大致可通过以下方法进行分类。
一、根据色素在不同溶剂中的溶解度差别进行除去
属于最常用、最简单、也是效果比较差的方法。
1.水提醇沉:可去除小部分水溶性色素。
醇提水沉:可除去大部分脂溶性色素。(也可以两种方法交替使用)
2.酸碱沉淀法:例如当杂质色素是一些黄酮、蒽醌等酚酸性成分时,可调节PH3以下,另其析出。
二、根据色素在两相溶剂中的分配比不同进行除去
例如当杂质色素是一些黄酮、蒽醌等酚酸性成分时,可采取调节PH到12以上,用有机溶剂萃取的方法。这时由于色素都以解离形式存在,不宜被萃出。
三、根据色素与有效成分吸附性差别进行分离
1.物理吸附:(吸附力是分子间力)
(1)极性吸附剂:如硅胶、氧化铝。可去除亲水性色素。
(2)非极性吸附剂:如活性炭,纸浆、滑石粉、硅藻土。可去除亲脂性色素。
活性炭是一种优良的吸附剂,它对色素、细菌、热原等杂质有很强的吸附能力,并且其还有助滤作用。
其内部有大量的微孔和空隙,表面积可达200-500m2/g。
吸附原理:由于大多数色素具有共扼双键结构,易吸附。
使用方法:冷吸附法,热吸附法,炭层助滤法,柱层析吸附法。
2.化学吸附:
(1)例如可用碱性氧化铝去除一些黄酮、蒽醌等酚酸性色素。
(2)离子交换树脂法:例如黄酮、蒽醌等酚酸性色素可以用阴离子交换树脂除去。
3.半化学吸附:聚酰胺与大孔树脂。
吸附原理为氢键作用,大孔树脂还有部分范德华力作用。
聚酰胺可通过分子中的酰胺羰基与酚类、黄酮类的酚羟基形成氢键。也可一通过酰胺键上的游离胺基与醌类、脂肪羧酸上的羰基形成氢键。
四、沉淀法除去色素
代表物质:石灰乳。
常用浓度:20%-30%。
脱色原理:石灰乳中钙离子能与药液中的有效成分及杂质结合成钙螯合物、钙盐沉淀。而沉淀在硫酸作用下,黄酮、蒽醌、酚类、皂苷、部分生物碱与钙离子形成的钙盐可以被分解出来,再溶解到水中。但是鞣质、部分蛋白质、有机酸、极性色素、多糖等不能分解出来。
五、絮凝剂法除去色素
1.常用的絮凝剂分以下几种:
(1)明胶类:鞣质影响药液稳定性且容易变色。可利用明胶与鞣质行政络合物,与水中悬浮颗粒一起沉淀
(2)ZTC1+1天然澄清剂:
分为四种:
I型:除蛋白型
II型:脱色澄清型
III型:中药口服液与颗粒剂型,可代替醇沉法,起到去除不稳定成分和助滤作用。
IV型:注射液型,主要提高澄明度。
(3)101果汁澄清剂
(4)甲壳素及壳聚糖:壳聚糖是甲壳素乙酰化制得。它们都是天然的阳离子絮凝剂。
2.影响澄清效果的因素
(1)澄清剂的用量
(2)澄清剂的配制浓度和加入顺序
(3)药液本身的浓度
(4)絮凝时温度的影响
(5)药液的PH的影响
(6)搅拌速度和搅拌时间的影响
(7)絮凝沉淀时间的影响
六、膜分离去除色素
最常用的为超滤技术。
